IL-36B binds to the IL1RL2/IL-36R receptor, activating NF-kappa-B and MAPK signaling pathways, which are linked to pro-inflammatory responses. It is part of the IL-36 signaling system and shares the coreceptor IL1RAP with the IL-1 family.
IL-36B stimulates the production of IL-6 and IL-8 in synovial fibroblasts, articular chondrocytes, and adipocytes. It also induces antimicrobial peptides, such as beta-defensins, and matrix metalloproteinases. In the skin, IL-36B acts on keratinocytes and dendritic cells, driving tissue infiltration and cell maturation, and promotes chemokine and pro-inflammatory cytokine expression, including TNF-alpha and IL-6.
